Prothrombin consumption, serum prothrombic activity and prothrombin conversion accelerator in hemophilia and thrombocytopenia.

چکیده
Attention has recently been focused on the consumption of prothrombin during blood coagulation. Studies of the velocity of prothrombin conversion to thrombin have yielded important information concerning the clotting defect in various hemorrhagic disorders. Thrombin evolution is retarded in the coagulation of hemophilic blood (1-5), thrombocytopenic blood (3, 4, 6, 7) and blood exposed to siliconized surfaces (8). This is reflected in the high residual prothrombic activity of the serum. Precisely how much prothrombin is consumed per unit of time is, however, not clear. Obviously, prothrombin consumption can be computed from the difference between the prothrombin concentration of serum and that of its parent plasma. The reliability of this computation rests upon the specificity of methods for measuring the concentration of prothrombin in both plasma and serum. The one-stage procedure measures the velocity of thrombin formation in the presence of optimal thromboplastin and calcium. In applying it to serum one must exclude the possibility that in the transition from plasma to serum substances arise which affect the velocity of prothrombin conversion. Convincing evidence exists, however, that during coagulation prothrombin conversion accelerators evolve which are demonstrable in serum (9, 10). Observations presented in this paper show that such an accelerator can be obtained from hemophilic, thrombocytopenic and silicone.8 sera and that it contributes toward their high one-stage

Recently, substances have been described which, arising in blood during its coagulation, accelerate the conversion of prothrombin to thrombin in the presence of thromboplastin plus calcium.3’ Their evolution and physiologic properties help cxplain ...

T HE prothrombic activity of plasma, as measured by the one-stage method, decreases as plasma ages (x-3) because of deterioration of ‘labile factor’ necessary for the rapid conversion of prothrombin to thrombin by thromboplastin plus calcium. When, however, determinations are made by a modified procedure in which the test plasma is first diluted with pdkombin-free fresh plasma, the aged plasma ...
متن کاملPREPARATION OF HIGHLY PURIFIED SOLVENTDETERGENT COA GULATION FACTOR VII AND FACTOR IX CONCENTRATES FROM PROTHROMBIN COMPLEX (PPSB)
In this study, anion-exchange chromatography was used to purify factor VII and factor IX from prothrombin complex (PPSB), which contains coagulation factors II, VII, IX and X. For this purpose, DEAE-Sepharose CL-6B gel , Pharmacia column XK-26 , high flow rate and two stepwise gradients with phosphate citrate buffer were used. The yield of the two lyophylized products, factor VII and factor...
متن کاملThe Prothrombin Conversion Accelerator of Serum
The conversion of prothrombin to thrombin during normal coagulation proceeds with ever increasing velocity until at the moment of clotting, or shortly thereafter, thrombin evolution is explosive. This phenomenon, referred to as the "autocatalytic" nature of thrombin formation, remains unexplained. It is evident that abnormalities in this reaction may underlie the pathogenesis of certain hemorrh...

By RICHARD WARREN, M.D. AND JOHN S. BELKO, M.S. A PRACTICAL CLINICAL METHOD for determining the activity in plasma of the accelerators of prothrombin conversion has not been so rapidly forthcoming as have been descriptions of their properties and reactions. Those methods which have been described have employed two general principles. The first. of these is the ability of the unknow’n plasma to ...
